This direct file download link works fine using the "Media" prefix...
[[Media:Wiki.png]] = Media:Wiki.png

...but I can't seem to make it work when defined in the "link" attribute of a file:
[[File:Download link.png|25px|link=Media:Wiki.png]] =

Is this expected? Is there any way to link directly to a downloadable asset from an image?

You should use [[File:Download link.png|60px|link=https://upload.wikimedia.org/wikipedia/mediawiki/b/bc/Wiki.png]]
